I'm using Nvidia gpu's.  I have the wallet but it is not fully synced yet.  I can cpu mine, but when I attempt to gpu mine I get this error:

[2017-07-03 19:07:53] HTTP request failed: The requested URL returned error: 500 Internal Server Error
[2017-07-03 19:07:53] json_rpc_call failed, retry after 30 seconds

over and over...

My .bat file is in my miner folder, which is in my spreadcoin folder with my wallet.dat file

The .bat file looks like this:

spreadminer -o http://127.0.0.1:41677 -u user -p password

pause

my spreadcoin.conf file looks like this:

miningprivkey=xxxxxxxxxxxxxxxxxxxxxxxx...
rpcuser=user
rpcpassword=password
rpcallowip=127.0.0.1
rpcport=41677
port=41678
gen=0
server=1

Do I need my wallet fully synced to gpu mine?

I followed the tutorial given on the Spreadcoin website, but that only acutely addressed AMD gpu mining.  The NVIDIA doesn't work in the same way.  I'm not sure what I'm doing wrong.

Yes, blockchain needs to be synched.

Error-Messages tell you that spreadminer can't connect with wallet.

Reason being that probably connection is blocked.

Firewall?
